autoinducer synthetase rhlI and GFP under PyeaR
The pathway detects nitrates in the medium; if KNO3 is present, the circuit is activated, RhlI produces the AHL (C4-homoserine lactone) and TetR is synthesized to block another pathway. GFP is emitted to show the start of transcription.
We test the operation of the promoter with different concentrations of KNO3, the data are shown above. We can see that the green fluorescence becomes stronger with higher amount of KNO3. That also means that the LasI and TetR are transcribed more efficiently with higher concentration of KNO3. When testing green fluorescence, we use 395nm as exictation, and detect green fluorensence with 509nm.
